RTA-408

RTA-408 (omaveloxolone) is a synthetic oleanane triterpenoid developed as a covalent Nrf2 pathway activator, structurally related to the CDDO family of compounds. In the literature it has been studied as a probe for Keap1-Nrf2 biology, oxidative-stress regulation, and mitochondrial function in cellular and rodent models. It is a triterpenoid, not a peptide — included in the catalog as a related research compound.

RTA-408 covalently modifies reactive cysteine residues — most prominently Cys151 — on Keap1, the substrate adapter that targets Nrf2 for proteasomal degradation. Modification of Keap1 stabilizes Nrf2, which translocates to the nucleus and binds antioxidant response elements (ARE) in promoter regions of phase II detoxification and antioxidant defense genes: HO-1, NQO1, glutathione synthetase, and GCLC/GCLM. Downstream effects on mitochondrial biogenesis and ATP production have been reported in cardiac and muscle cell models.

Used in oxidative-stress and mitochondrial-function research, including studies on Friedreich ataxia models, neuronal protection assays, and ischemia-reperfusion injury models in rodent and cell-culture systems.
